Londonderry unveils city of culture program

(AP) ? The Northern Ireland city of Londonderry has announced a program of arts events ranging from punk to painting to celebrate its role as the U.K.'s "city of culture" for 2013.

The program announced Thursday includes a new play by U.S. writer Sam Shepard staged by Field Day, the innovative local company founded in 1980 by playwright Brian Friel and actor Stephen Rea.

Other stage offerings are "Teenage Kicks," a punk musical inspired by local rock heroes The Undertones, and an open-air spectacle devised by Frank Cottrell Boyce, who scripted the opening ceremony of the London Olympics.

Nobel Literature prize-winning poet Seamus Heaney, who comes from Northern Ireland, will kick off a poetry festival in the city, which will also host an exhibition for art's Turner Prize, performances by the London Symphony Orchestra and a visit from the Royal Ballet.

The U.K. city of culture title is inspired by the continent-wide European capital of culture program. Londonderry is the first community to hold the title.

Organizers hope it will give an economic boost to Northern Ireland's second city, which is scarred by sectarian divisions that even extend to its name ? Protestants call it Londonderry, Roman Catholics Derry.

The city has been targeted by IRA dissidents in a series of shooting and bombing attacks, including an incident in which a small bomb in a backpack was left outside the City of Culture offices.

Despite this, Northern Ireland's peace process has helped transform the city's fortunes. It was recently named one of the best cities in the world to visit next year by the Lonely Planet travel guides.

Garth Brooks Has Family's Blessing to Resume Career - The Boot

Mark Humphrey, AP

Garth Brooks is one of the latest members to be inducted into the Country Music Hall of Fame, which may be the pinnacle but by no means marks the end of his illustrious career. The singer, whose three-year run as a performer at the Wynn Las Vegas ends next month, hints that fans may hear more music from him in the near future ... with his family's blessing, of course.

"We're just going to start," Garth tells The Huffington Post. "It will be fun. We've got our youngest baby [Allie], this is her junior year, and she's really in touch. Now, down from three to one [at home], you spend a lot more time with that one. She's in on every plan. She knows all the decisions. She's really well-educated in what's going on."

The Oklahoma native previously said he would consider resurrecting his music career when all three daughters were grown and out of the house, but his wife, singer Trisha Yearwood, reveals that there's another Brooks family member who would like to see that happen sooner.

"I think she's ready for us to go on the road because she'll be home by herself," Trisha jokes of her youngest stepdaughter. "She's doing every after-school activity you can do. 'You don't just want to sit and stare at us?' It's really fun."

Garth says he is counting on several of his fellow musicians to help him ease into the next chapter of his career. "You talk about songwriters, the great ones can birth a career but the greatest ones can bring life back into one," he notes. "So we'll be looking hard for songs just to see what the future might hold for us."

For now, Garth has four more shows to play in Sin City. See his remaining Wynn Las Vegas dates here.

Seagate Backup Plus brings USB 3.0 speed, sociability to new Macbooks

DNP Seagate Backup Plus spices up Mac offerings with USB 30

When Seagate launched its PC/Mac Backup Plus offering that could safeguard not only your folders but your Facebook or Twitter content, too, it seemed like a nice idea. But conspicuously missing from the Mac side of that equation was a USB 3.0 port to hustle transfers along at a much less pedestrian 5 Gbps -- likely because until recently, no Macs directly supported it. Now, Seagate has launched a USB 3.0 version of the device for those shiny new Macbook Pro Retina and Air models that pack it, while offering the option to upgrade to Thunderbolt or FireWire 800 "as the need presents itself." Prices go from $110 for the 500GB model up to $180 for the 3TB version -- check the PR after the break to see the entire range.

Roku's New Streaming Stick for TVs Lets You Ditch the Set-Top Box ...

According to Forrester Research, more than 32 million U.S. consumers are now using an assortment of devices to watch Web video on their TV sets, from gaming consoles to set-top boxes like Apple TV, Roku and Google TV. These are often considered interim devices, while ?smart? TVs ?- ones that are always Internet-connected ?- ease their way onto the market and into living rooms.

But adding these boxes to your TV setup means factoring in more wires and easy-to-lose remote controls. And finding the right input for the device through the TV remote can leave even the smartest people feeling dumbfounded.

Roku, the California-based company that makes set-top boxes of the same name, has come up with a new solution to this: A wireless stick, not much bigger than a thumb drive, that plugs directly into the back of your TV to stream HD video from the Web ? turning your ?dumb? TV into a smart one, with minimal gadgetry.

The $100 Roku Streaming Stick, available through Roku.com or Amazon.com, offers the same Web apps and the same powerful processor as Roku?s top-of-the-line set-top box, the Roku XS 2 (also $100). This means that all of that streaming goodness is packed into a small stick.

The Roku Streaming Stick also responds to the same remote used for your TV, as well as to a remote control app for iPhone and Android smartphones ? eliminating the need for at least one extra remote.

But there?s one major catch with the Roku Streaming Stick: It only works with a certain type of TV set, one that includes a port called an MHL (Mobile High-Definition Link) port.

While Roku?s CEO Anthony Wood says that the company expects to see more of these TVs on the market within the next year, there are currently around 50 sets and displays sold in the U.S. that are MHL-equipped. And, of those, Roku has certified only a few brands that work really well with the device: Insignia, Apex and Hitachi.

In order to test the stick, I had to shift my regular TV set to the floor and set up a television that Roku delivered to me: a 55-inch, HD Insignia TV that costs $900. Normally, I wouldn?t replace my existing TV with another one just because I wanted to attach a $100 gadget to it, and I?m guessing a lot of consumers would feel the same.

But the plug-and-play stick is convenient. It?s a few inches long and weighs just three ounces. It?s bright purple, and plugs directly into a color-coded MHL port on the backside of MHL TV sets.

It has no wires, and one of the benefits of this technology is that the MHL ports actually charge the product while it?s plugged in, so there?s no need to charge the stick at any point. The set-up took all of 10 minutes, once I plugged the stick into the TV, authenticated the device and registered with Roku on the Web.

Roku says it has more than 500 apps, which it calls ?channels,? on its platform, including popular apps like Netflix, MLB.tv, Hulu Plus, Amazon Instant Video, Vudu and Facebook. Both the Roku XS 2 box and the stick come with Angry Birds, which can be played using Roku?s motion-sensor remote.

Over the past week, I?ve watched a lot of shows and movies through the Roku Streaming Stick, mostly through Netflix, Amazon and Vudu (the last two charge per movie rental or download, unlike Netflix?s monthly subscription). I also listened to NPR radio, streamed music through Pandora, and logged into Roku?s bare-bones Facebook app, which lets users view photos and videos, but doesn?t allow for status updates.

The media-streaming quality was very good. Occasionally, HD movies would pause to buffer, but never for longer than a few seconds. Only one movie channel I checked out, Sony?s Crackle channel, paused consistently, to play short ads. This was annoying, but then again, the Crackle movies were free.

One app missing from Roku?s offerings is YouTube. I?ve spent more hours than I care to admit using Apple TV to watch YouTube videos with friends. Roku also doesn?t have a Web browser. And this might seem obvious, but you can?t watch media you?ve purchased through iTunes or the Google app store on Roku?s device, either.

Lastly, cable-authentication apps that require a user to have a cable subscrition ? like HBOGo or Epix ? will only work on Roku with some cable providers. For example, DirectTV and Comcast do not support HBOGo on Roku. Time Warner Cable, however, does.

As with other set-top boxes, text input is clumsy: You use the arrow keys on the remote to select individual letters of the alphabet. But you don?t actually need the Roku Streaming Stick?s own remote unless you want to play Angry Birds or other games that use motion control. The Stick is compatible with the MHL TV?s remote, so I could access and navigate Roku by pressing the ?Home? button on the Insignia TV?s remote and using those arrow buttons.

Being able to just press ?Home? to get to the Roku stick was joyfully simple compared to the usual process of searching my TV inputs for different devices I?ve connected to it.

Roku also offers new iOS and Android apps that control the Roku Streaming Stick. Much like Apple?s AirPlay feature, I could use the Roku mobile app to stream the many photos stored on my iPhone directly to the TV set, provided that both devices were connected to the same Wi-Fi network.

The Roku Streaming Stick successfully compresses the experience of a Web video box into a tiny, convenient device, but it comes with a lot of ?ifs.? Even Roku says that its Stick may eventually be sold as a bundle rather than as a singular gadget. If you prefer Roku?s channel offerings over other set-top boxes, if you have an MHL-compatible television set, and if you?re not planning on upgrading to a high-end smart TV with built-in Internet capabilities anytime soon, it?s a great device. Otherwise, it?s a niche product right now.

Book Review - Create with Maisy: A Maisy First Arts-and-Crafts Book

About the Book
Let Maisy show you how! Easy, safe, and fun, these craft projects for the very young were put together and photographed by Lucy Cousins herself. Why does Maisy keep a Big Box of Everything, filled with glitter, sequins, foil, stickers, paint, brushes, paper, and tons more stuff? Because she loves to make things! Children as young as two will be thrilled to follow her lead, thanks to a brand-new Maisy format featuring both familiar illustrations and photographs by Lucy Cousins. Here are crystal-clear steps for creating seventeen simple crafts, highlighted by photos of the end result. Enjoy making things from painted pebbles to paper lanterns, food collages to feathered masks, and wax-relief paintings to wonderful vegetable animals. As spectacular looking as it is hands-on, here is an activity book sure to please parents as much as it does Maisy?s little fans.

My Take on the Book
If you love to work on projects with your child at home or if you are searching for the perfect gift for a child's birthday or holiday which utilizes their love of arts and crafts, you will love this book by Lucy Cousins. Lucy's book if full of easy to make projects and most of them use materials you may already have around your house. If you don't currently have the materials, they could easily be found at any art and craft store or other store which carries craft materials.

The book is child centered with easy to follow directions and illustrations for either step by step or/and the finished project. This book holds hours of creative fun for you and your special child.

As a gift , my thought was it could be given with a bag full of supplies so that the child can begin to make the projects. Just think of the joy you would bring to a child who loves to create with such a useful gift.

I encourage you to seriously think about this book as one to keep as a resource for those days you are looking for a perfect activity indoors.

Germany likely posted surprise growth in Q3: finmin report

BERLIN (Reuters) - The German economy likely grew in the third quarter despite a weak global backdrop thanks to a strong performance by German industry and robust exports to areas outside the euro zone, the Finance Ministry wrote in its monthly report.

"Expansion in German industrial production...likely gave significant growth impetus. Most analysts did not expect this given the deterioration in the business mood," the report said.

Many economists had forecast the economy would contract in the third quarter, particularly as Germany's closely-watched Ifo business sentiment index worsened for five months in succession.

"All in all, German exports continue to appear robust despite a slow-down in global momentum... Stimulus for the export market came above all from non-euro zone countries," the report said.

German growth slowed to 0.3 percent in the second quarter from 0.5 percent in the first as firms postponed investments due to concerns about the 17-nation bloc's troubles.

The Finance Ministry said in the fourth quarter of 2012 there could be a significant weakening in economic activity, due to the depressed business climate in other euro zone states, but the German economy would pick up gradually during 2013.

Last week the government slashed its 2013 growth forecast to 1 percent from 1.6 percent due to European economic weakness, though it lifted its expectation for 2012 by 0.1 percentage points to 0.8 percent.

Germany's economy powered through the first two years of the euro zone's sovereign debt crisis, posting 4.2 percent growth in 2010 and 3 percent last year at a time when some peers were seeking bailouts and others were grinding to a halt.

However the woes of some of its southern European partners have now started to impact economic performance.

Jonathan Carbutti - New Haven Real Estate Expert: Who Said ...

Watch on your mobile device >>

Other than the age-old trend of people buying homes more so in the spring season as opposed to the fall and winter, there really is no reason that one might choose one season over the other. Sure, there may be more homes on the market ? but equally there are as many more buyers circuiting the market too.

The question prospective buyers ought to be asking themselves is whether or not they would prefer buying a home that is right for them regardless of the time of year or does it just suit them more to buy when it?s warmer outside?

Spring Is In the Air ? And So Is Much More Competition

Many buyers like to be out shopping for a new home at a time that they feel there are a lot more homes on the market. But one very important thing to consider is that there are equally as many other buyers out there also looking. And with a greater number of buyers at any given time, there?s a good chance you will come across more buyers looking for the same type of property that you are seeking.

So when the perfect home shows up on the market, in great condition and priced just right ? you guessed it ? more than one buyer will likely show up to make an offer on it. What does that do to your chances of securing the home of your dreams? Having multiple buyers vying for the same property dilutes your chances a good amount. Not only will you find a greater likelihood of competing against another buyers? bid but you will also face the risk of multiple offers and ultimately a bidding war. Despite the low, low interest rates, the last thing any buyer wants is to deal with artificially inflated prices.

Real Estate Market in Fall and Winter Slows Down But Doesn?t Stop

Contrary to what many people believe, the real estate market does not stop during the cooler months of the year ? rather it just slows down a little. But this can work in buyers? favor when you consider that there is fewer buying competition to contend with. Yes there are fewer homes on the market since the same people that believe springtime is a better time for real estate also remove their homes from the market. However, with fewer sellers there are also fewer buyers which means that when you do find the perfect home there is a much better chance at successfully closing the deal ? and without all the hoopla that goes along with multiple offers or bidding wars.

Consider also that by the time the holidays begin to roll around, many sellers have had their property on the market for one, two or even more months and they are eager to sell. This translates to their willingness to negotiate more than ever before.
~
With today?s record setting low interest rates and with them the very low cost of borrowing money combined with increased buying power, it only makes sense to jump off the fence and embark on your home buying journey. We invite you to contact us today so we can begin helping you find your perfect home.

Nurses, cleaners, librarians: UK austerity ... - World News - MSNBC

Dan Kitwood / Getty Images

A man holds up a banner reading 'Austerity - That's Enough' as a march to protest the government's austerity measures prepares to set off from the Embankment in London on Friday.

By NBC News staff and wires reports

Updated at 8:25 a.m ET: LONDON - Thousands of anti-austerity protesters marched in London on Saturday to protest against public spending cuts enacted by a government fighting off accusations that it is run by an upper-class elite that ignores the plight of recession-hit voters.?

The march comes at a time when Prime Minister David Cameron's Conservative-led coalition is reeling from the resignation on Friday of a senior minister accused of calling police "plebs," a class-laden insult for working people.?

Conservatives faced a barrage of negative headlines on Saturday over the departure of Andrew Mitchell, the "Chief Whip" or party enforcer, four weeks after he swore at police guarding the gates to Cameron's Downing Street office.?

Class wars: 'Gate-gate' scandal swamps UK PM David Cameron

A second row involving George Osborne, the finance minister -- who sat in a first class train carriage with a standard class ticket before paying for an upgrade -- played into the hands of critics who say the Conservatives are privileged and out-of-touch.?

"Who Do They Think They Are?" asked the Daily Mail newspaper in a front page headline, while the Financial Times said the bad news over Mitchell and Osborne capped a "dismal week for the Tories", the center-right party that is trailing in the polls.?

Nurses, cleaners, librarians and ambulance drivers are among tens of thousands marching past the Houses of Parliament to a rally in Hyde Park in one of the biggest anti-austerity protests this year. Marches will also take place in Belfast, Northern Ireland, and Glasgow, Scotland.?

General strike?
Under grey autumnal skies, police closed roads around parliament in Westminster before the start of the march at 1100 GMT (6 a.m. ET).?

Trade union leaders are seeking to pile more pressure on Cameron at the event where they will tell protesters that the government's economic plan has failed, prolonging Britain's second recession since the financial crisis.?

Greek strike turns violent: 'Enough is enough'

The head of the head of the RMT transport workers union was set to say that Saturday's march was a step towards a nationwide strike, Sky News reported.

"The marches are a building block towards the objective of coordinated action and a general strike. ?That is why RMT says, march today, strike tomorrow,"?Bob Crow was planning to say, Sky News reported without citing a source.

Reuters reported that Brendan Barber, head of the Trades Union Congress, an umbrella group which represents 54 unions, planned to say the following:

"Austerity isn't working. It is hammering the poorest and the most vulnerable ...?Ministers told us that if we only accept the pain, recovery would come. Instead we have been mired in a double-dip recession."?

Coalition under pressure
The coalition government has responded to calls from unions and the opposition Labour Party to do more to boost growth by relaxing planning laws and boosting lending to businesses.?

A downsized parade as Spain's celebrates national day amidst austerity cuts

But its latest attempt to ease the pressure on squeezed households backfired this week when Cameron said the government would legislate to force energy companies to give customers their lowest tariff. The surprise announcement appeared to take his own ministers by surprise and sowed confusion over what he meant and whether it would actually happen.?

However, Sajid Javid, a Conservative Treasury minister, said the government was right to focus on cutting borrowing and that data last week indicating a fall in unemployment and inflation showed that its economic policies were on track.?

"There is a still a lot to do," he told Sky News. "I don't pretend for a second that we are out of the woods, but this government is facing up to the problem, it is not sticking its head in the sand like (Labour opposition leader) Ed Miliband."?

Asked about the perception that the Conservatives are out of touch, he said: "I think that what matters is what is actually happening out there in the real world."?

Spain, Portugal hit with anti-austerity protests

Opponents of the unions say the government should stick to its plan to eliminate a budget deficit that stood at 8 percent of gross domestic product last year, the biggest of any major European country.?

"The government must not listen to militant union leaders," said Mark Littlewood, director general of the Institute of Economic Affairs, which describes itself as an independent free-market thinktank. "The cuts we have seen are tiny and further concessions to these protesters would be wholly unaffordable."?

Reuters contributed to this report.

Riders stranded aboard Stratosphere at Texas State Fair

Just hours after "Big Tex" burned to the ground, the Stratosphere strands riders high above the Texas State Fair.

By NBCDFW.com staff

Updated at 12:37 a.m. ET: One of the State Fair of Texas' tallest rides malfunctioned Friday night, stranding two dozen riders in the air for more than two hours.

Riders in the Stratosphere's hanging seats were stuck dangling about 165 feet in air when the ride lost power at about 9:11 p.m.

Carol Stradtman, one of the stranded riders, told NBC 5 by phone that all of the lights went off and the ride just stopped.

"It's extremely scary," she said.

View NBCDFW.com for complete coverage

The 24 riders were on the ground at about 11:20 p.m. Dallas Fire-Rescue said all of the occupants were safely removed from the ride after it was manually lowered.

Power was restored to the ride by 10:50 p.m., but there appeared to be some issue that prevented the ride from immediately lowering its passengers.

Two Dallas Fire-Rescue technical rescue trucks were at the fair to help.

Stradtman told NBC 5 that someone had climbed the center of the tower at about 9:40 p.m. and appeared to manually?attempt to bring them down. She said a friend who was not on the ride said there was some kind of mechanical difficulty but said she had not been given an update on when or how they would get off the ride.

No injuries have been reported, Dallas police said.

The Stratosphere, a 200-foot tall tower, turns and swings riders out over the fair.

The Dutch-made ride stalled at the Minnesota State Fair twice in as many days in August, the Minneapolis Star Tribune reported.

NBC 5's Ray Villeda and Ellen Goldberg contributed to this report.

Ivy Tech, Humane Society accelerate animal shelter dialogue ...

TERRE HAUTE ? Talks continue between Ivy Tech Community College and the Terre Haute Humane Society to locate the animal shelter at the former Doughmakers building, with Ivy Tech offering a veterinary technician and related programs.

?We feel very confident at this point? that it will happen, said Ann Valentine, chancellor at Ivy Tech?s Wabash Valley region. The proposal first became public in January.

Valentine said Ivy Tech has signed a memorandum of understanding with the Humane Society board. Both will help fund a? feasibility study to determine the success of a fundraising campaign to support the collaborative project.

A joint fundraising campaign would look to raise $2 million to $4 million, Valentine said.

Part of it, $200,000, would be used for Ivy Tech to purchase equipment for new programs, including one that trains veterinary technicians. Other possible programs include animal-assisted therapy, kennel management, and grooming.

The campaign also would pay for renovation to part of the Ivy Tech Learning Achievement Building (TechLAB), formerly known as Doughmakers, to house the animal shelter. The shelter would have to have a separate air handling system from the building?s nursing, and potential diesel technology, programs.

The shelter also would need such things as additional kennels, cages and a commercial laundry system, Valentine said.

The humane shelter would be located on the west side of the TechLab building, she said. That way, the shelter could open kennels and runs toward grass and trees on the west side and away from U.S. 41.

?Our hope is if we could get as far as $4 million [in a campaign], we could endow some of their operating expenses,? Valentine said

Depending on how the feasibility study goes, a fundraising campaign could start early next year. Initial feedback ?has been pretty positive,? she said.

Ivy Tech would like to begin offering some of its new programs next fall. Starting those programs would not be contingent on the animal shelter locating there, if more time was needed to raise sufficient funds.

The proposal to locate the shelter there ?is very much alive and we?re making progress,? Valentine said.

Debbie Floyd, board president of the Humane Society, also is optimistic. ?We?re definitely making progress and working toward it.?

?She noted that building plans have been drawn up, although they need to be fine-tuned.

?Every indication I have is that this is a wonderful opportunity for Ivy Tech, the Humane Society and the community,? Floyd said. ?The building we have is not a healthy environment for this day and age.?

The new location, with the services that Ivy Tech programs could provide, ?will help us improve ourselves,? Floyd said.

Study questions feasibility of entire genome sequencing in minutes

ScienceDaily (Oct. 17, 2012) ? The claim that nanopore technology is on the verge of making DNA analysis so fast and cheap that a person's entire genome could be sequenced in just minutes and at a fraction of the cost of available commercial methods, has resulted in overwhelming academic, industrial, and global interest. But a review by Northeastern University physicist Meni Wanunu, published in a special issue on nanopore sequencing in Physics of Life Reviews, questions whether the remaining technical hurdles can be overcome to create a workable, easily produced commercial device.

Earlier this year Oxford Nanopore Technologies, one of the pioneering companies of sequencing discoveries, announced that they expect nanopore strand sequencing to achieve a 15-minute genome by 2014 at a cost of $1,500. This is a far cry from the $10 million it cost to sequence an entire genome just 5 years ago.

Since the idea of nanopore sequencing was first proposed in the mid 1990s, huge advances have been made. The basic idea is exceedingly simple: a single thread of DNA is passed through a tiny molecule-sized hole-or nanopore-and the various DNA bases are identified in sequence as they move through the pore.

But according to Wanunu, the reality of manipulating technology based on pores so tiny that 25,000 of them can fit side by side on a human hair has proved a daunting task. The main challenge has been to slow the process down and control the movement of the DNA strand through the pore at a rate slow enough to make individual DNA bases readable and usable. A new approach using enzyme-controlled movement, developed to overcome this problem, has its own drawbacks including poor enzyme activity resulting in limited processivity and uncontrolled forward-reverse motion.

Another major dilemma has been whether protein or solid-state pores provide the most promising technique. At first, naturally occurring porous proteins were investigated. But in the early 2000s, heralded as offering better capability and flexibility, various solid-state nanopores made of silicon or graphene were tested. "Since both lipid-embedded protein channels and solid-state nanopores have drawbacks, it will be interesting to see which device, or what combination of devices, will be available in years to come, if any," Wanunu says.

At this time there are still many hurdles to overcome, he adds, including the inability of nanopores to provide any spectroscopic information about the identity of a molecule, uncertainties about whether translocation occurs at a constant speed, and the complications of pore clogging.

Writing in a comment published in the same issue, John Kasianowicz from the National Institute of Standards and Technology in the US, a pioneer in the field, agrees that plenty of challenges remain: "There are indeed still many problems to address in order to enable practical electronic nanopore-based sensing devices. However, by better understanding the road already developed in this nascent field, the journey will hopefully appear a little less daunting."

In a final comment on Wanunu's review, the founder and Director of Oxford Nanopore, Hagan Bayley, looks ahead to the future: "In the longer term, by using solid-state pores... it may be possible to read DNA sequences at microseconds rather than milliseconds per base. This could be done by using tunnelling currents or other characteristics of the DNA bases for which graphene-with its unusual electronic properties-might after additional development provide a superior substrate and in so doing deliver another massive leap forward on top of a decade of unprecedented progress."

Many genes, but two major roads to adaptation ? Nothing in biology ...

Cross-posted at Denim and Tweed.

In the course of adaptive evolution ? evolutionary change via natural selection ? gene variants that increase the odds of survival and reproduction become more common in a population as a whole. When we?re only talking about a single gene variant with a strong beneficial effect, that makes for a pretty simple picture: the beneficial variant becomes more and more common with each generation, until everyone in the population carries it, and it?s ?fixed.? But when many genes are involved in adaptation, the picture isn?t so simple.

This is because the more genes there are contributing to a trait, the more the trait behaves like a quantitative, not a Mendelian, feature. That is, instead of being a simple question of whether or not an individual has the more useful variant, or allele, at a single gene ? like a light switch turned on or off ? it becomes possible to add up to the same trait value with different combinations of variants at completely different genes. As a result, advantageous alleles may never become completely fixed in the course of an adaptive evolutionary response to, say, changing environmental conditions.

That principle is uniquely well illustrated by a paper published in the most recent issue of Molecular Ecology, which pairs classic experimental evolution of the fruitfly Drosophila melanogaster with modern high-throughput sequencing to directly observe changes in gene variant frequencies during the course of adaptive evolution. It clearly demonstrates that when many genes contribute to adaptation, fixation is no longer inevitable, or even necessary.

Turning up the heat, homogenizing flies

The authors of the new study, a team from the Institut f?r Populationsgenetik led by Pablo Orozco-terWengel, conducted what would otherwise be a rather simple experiment in evolutionary change in the laboratory. Starting with fruitflies collected from a wild population in Portugal (yes, Virginia, Drosophila melanogaster has wild populations!) they established three replicate populations of about 1,000 flies, which they put in temperature-controlled conditions somewhat warmer than the original collection location, and allowed them to propagate for 37 generations. Exensive previous work with Drosophila has established that simply moving the flies into a laboratory setting ? where they live in bottles, and eat prepared food ? exerts natural selection on them, and the increased temperature added a little bit more novelty to the lab environment to make it more likely adaptation would occur.

This experiment is different from all that previous experimental evolution of Drosophila, though, is that the coauthors tracked allele frequencies at thousands of markers during the course of those 37 generations of adaptation to the lab. To do this efficiently, they used an approach called ?pooled sequencing.?

The principle behind pooled sequencing is that, if all you care about is the relative frequency of a gene variant in a whole population, you don?t need to know the genotype of any specific individual in that population. So to track changes in allele frequency, the team sampled hundreds of flies from the experimental population, and ground them all up together. (The polite, technical term used here is ?homogenized.?) They then extracted DNA from this ?pooled? sample, and used a high-throughput sequencer to collect millions of reads ? short snippets of DNA sequence ? out of the pool as a whole.

To extract allele frequencies from all of those sequence reads, the team identified where each read matched the Drosophila melanogaster reference genome. When multiple reads matched to the same location, but differed in one or more DNA nucleotide bases, they identified those bases as variable markers ? single-nucleotide polymorphisms, or SNPs. Because the original DNA sample was pooled from many mashed-together flies, the relative frequency of each different variant of a SNP in the Illumina output should reflect the relative frequency of that SNP variant in the population as a whole.

Using this approach, Orozco-terWengel et al. could track allele frequency changes across more than a million SNP markers by taking these pooled samples from the intial population of flies, then at multiple points during the 37-generation evolutionary experiment. By comparing the allele frequencies in samples taken during the course of adaptation to the allele frequencies in the sample from the starting population, they could identify SNPs that became more common as the population adapted ? and, because they had a big sample from across the genome, they could identify those SNPs whose allele frequencies had changed more than would be expected due to genetic drift. They examined samples taken after 15 and 27 generations of evolution, and at the end of the 37-generation experiment.

Two paths to adaptation

Allele frequency changes (AFC) in SNPs showing significant change by generation 15 (a) and by generation 37 (b). Image from Orozco-terWengel et al. (2012), figure 3.

What they found was largely in line with the verbal model I outlined at the beginning of this post. Over the course of experimental evolution, significant increases in allele frequency occurred at thousands of SNPs ? suggesting that a great many genes are involved in the process of adaptation to life in the lab. Accordingly, very few of those allele frequency changes (in about 0.5% of the 2,000 SNPs that showed the greatest change from start to finish) represented complete or near-complete fixation.

More interestingly, comparison of allele frequency changes at the 15th generation and at the end of the experiment revealed two major ?paths? taken by alleles. In the first case, the SNPs with strongest allele frequency changes by generation 15 all hit a ?plateau? in subsequent generations ? they didn?t see any significant increase in frequency between generations 15 and 37. In the second case, SNPs with the strongest allele frequency changes by generation 37, the end of the experiment, had increased steadily from the beginning population through the samples taken at the 15th and 27th generation. The SNPs in this second set had not shown significant allele frequency increases by generation 15 ? which means the SNPs underlying most of the adaptive change in the first half of the experiment were a completely different set than the SNPs underlying adaptive change in subsequent generations.

If it?s already adapted, don?t fix it.

On the one hand, that suggests that Orozco-terWengel et al. managed to capture SNPs with a range of different contributions to the adaptation the observed by the end of the experiment. The SNPs with the biggest contribution showed rapid initial increases in allele frequency, then leveled off; SNPs with weaker effects showed slower, steady increases that continued for the entire experiment. But if it?s that simple, why didn?t the large-effect SNPs show continuing allele frequency change after the midpoint of the experiment?

It may be, as the coauthors speculate, that the two classes of SNPs identified in their experiment are separated by more than just the size of their respective contributions to adaptive change. There could be interactions among the alleles at these SNPs, such as overdominance, in which an individual is most fit when he or she carries two different alleles at a locus, rather than two copies of either allele. Overdominance would explain why most of the SNPs showing rapid initial increases in allele frequency then leveled out at intermediate frequencies.

So this combination of experimental evolution and modern sequencing technology raises some interesting questions even as it supports a lot of previous thinking about how natural selection acts on traits that are created by the collective action of many genes. It?s an exciting result, and, I hope, inspiration for much more work digging into the details of such ?polygenic? adaptation.?

Forward-Thinking Family House with Volumes Integrated

Play of Volumes Forward Thinking Family House with Volumes Integrated

Masahiko Sato, the architect of Architect Show?has defined the K5 house, in Kurume, Japan. With a futuristic kind of look and incredible high ceilings in one of the wings, K5 is ideal for young families, who are not afraid to embrace asymmetry and volumes, without questioning the architect?s sanity. In contrast to the rural landscape, on Sato?s project is written ? avant-garde? all over. Amazingly young spirited, the house juggles with contrasts of black and white and creates an impact on whoever ends up seeing it. Different angles, the irregularities ?in terms of volumes, every single element was designed to blend into a forward- thinking type of dwelling.

Contrasting Black and White Forward Thinking Family House with Volumes Integrated

Despite of its daring design, the place is really welcoming. By using natural wood as a main material in defining the interior, especially in the opened space kitchen- dining room area, the architect created a cozy atmosphere, ideal for family gatherings. The Western inspired interior is combined with elements of Japanese style, in order to fit the Far Eastern tradition. Due to the Japanese culture, a tatami room has been smoothly integrated into the project. The house?s irregular shape was mainly possible because of the peaceful location. A crowded urban site could not sustain under no circumstance such spatial variations. Very interesting and inspiring, K5 brings together the Western and the Eastern cultures, creating a perfect blend of style for modern families.

Re: How far have you ridden?

Postby MarcS ? Tue Oct 16, 2012 12:17 pm

That's pretty hard-core guys. I don't think my backside could possibly stand 60 miles of riding in one day--even if were on a flat, smooth road! The furthest distance I've ridden all at once on nothing but singletrack is right around 30 miles. I'm using the term "singletrack" a little loosley here. The trail I rode that day is a National Scenic Trail in the Oregon Cascades. None of the recreation maps of the area indicate that it's rideable; and the sign at the trailhead, although not prohibiting bikes, does warn would-be riders that it is not suitable for MTB. I just had to try, though. Needless to say, it was a gruelling ride over ridiculously rocky, steep terrain. Ironically, the thing that made such a long (for me) ride tolerable was that I was forced to be on my feet as much if not more than on the saddle.

25 primate species reported on brink of extinction

NEW DELHI (AP) ? Twenty-five species of monkeys, langurs, lemurs and gorillas are on the brink of extinction and need global action to protect them from increasing deforestation and illegal trafficking, researchers said Monday.

Six of the severely threatened species live in the island nation of Madagascar, off southeast Africa. Five more from mainland Africa, five from South America and nine species in Asia are among those listed as most threatened.

The report by the International Union for Conservation of Nature was released at the United Nations' Convention on Biological Diversity being held in the southern Indian city of Hyderabad.

Primates, mankind's closest living relatives, contribute to the ecosystem by dispersing seeds and maintaining forest diversity.

Conservation efforts have helped several species of primates that are no longer listed as endangered, said the report, prepared every two years by some of the world's leading primate experts.

The report, which counts species and subspecies of primates across the world, noted that Madagascar's lemurs are severely threatened by habitat destruction and illegal hunting, which has accelerated dramatically since the change of power in the country in 2009.

Among the most severely hit was the northern sportive lemur, with only 19 known individuals left in the wild in Madagascar.

"Lemurs are now one of the world's most endangered groups of mammals, after more than three years of political crisis and a lack of effective enforcement in their home country, Madagascar," said Christoph Schwitzer of the Bristol Conservation and Science Foundation, one of the groups involved in the study.

"A similar crisis is happening in Southeast Asia, where trade in wildlife is bringing many primates very close to extinction," Schwitzer said.

More than half of the world's 633 types of primates are in danger of becoming extinct because of human activity such as the burning and clearing of tropical forests, the hunting of primates for food and the illegal wildlife trade.

While the situation appears dire for some species, wildlife researchers say conservation efforts are beginning to pay off, with several primates being removed from the list, now in its seventh edition.

India's lion-tailed macaque and Madagascar's greater bamboo lemur have been taken off the endangered inventory for 2012 after the targeted species appeared to have recovered.

Also, conservation efforts have ensured that the world did not lose a single primate species to extinction in the 20th century, and no primate has been declared extinct so far this century, said Russell A. Mittermeier, president of Conservation International and the chairman of the IUCN Species Survival Commission's primate specialist group.

"Amazingly, we continue to discover new species every year since 2000," Mittermeier said. "What is more, primates are increasingly becoming a major ecotourism attraction, and primate-watching is growing in interest."

In a separate report on global urbanization released Monday at the Hyderabad conference, the United Nations urged urban planners to incorporate green spaces in cities as more and more people move away from rural areas in search of work.

Green areas in big cities perform important ecological functions, such as "filtering dust, absorbing carbon dioxide from the air and improving air quality," the Convention on Biological Diversity said in its new assessment.

The "Cities and Biodiversity Outlook" is the first global analysis of how urban land expansion will impact biodiversity in the coming decades.

The world's total urban area is expected to triple between 2000 and 2030, with urban populations set to double to around 4.9 billion in the same period.

Data from the United Kingdom show that a 10 percent increase in tree canopy cover in cities may result in a 3-4 degree Celsius decrease in ambient temperature, thus reducing energy used for air conditioning, the report said.

Urban biodiversity also delivers important health benefits, with studies showing that proximity to trees can reduce the prevalence of childhood asthma and allergies.
